The reason the two regions look different even though they are the same color is because of lateral inhibition. It's the way your visual system does edge detection. The photoreceptors in your eyes are wired together in such a way that the input to one receptor affects the nearby receptors because it affects how much one photoreceptor inhibits the ones around it. Essentially, an object's surrounding affect how your perceive it.

Because square B is surrounded by darker squares as well as the shadow from the pillar, the input to the eyes from square B is inhibited less. This is why it appears to be white. However, square A is surrounded by lighter squares and since that causes the signals from square A to be inhibited more which is why they appear to be darker.

And when _leech_ blacks out everything except the two squares, the visual signals are both inhibited the same amount, which is why they now appear to be the same.
